Currently, regular cow's milk contains 2 main types of protein: A1 protein and A2 protein. In the past, all cows were A2 cows. Over time, as a result of mutations, cows began to produce milk with A1 protein. Studies show that only 30% of cows still contain 100% A2 protein. Milk contains various proteins known as caseins and whey proteins. The caseins in A1 and A2 milk differ at a single position in the chain, by the presence of different amino acids. This variation is crucial because, unlike A1 milk, A2 milk does not produce casomorphin 7 (BCM7) during digestion. BCM7 is the result of the breakdown of proteins, and is thought to have harmful effects on the body.

For Holle A2 powdered milk formulas, only milk from specially selected cows is used, which do not contain A1 protein, but only A2 protein. The selection is based on high-performance DNA tests.
